Abstract: Candida albicans is a common cause of nosocomial infections whose virulence depends on the reversible switch from blastoconidia to hyphal forms. Neutrophils (or polymorphonuclear leukocytes (PMNs)) readily clear blastoconidia by phagocytosis, but filaments are too long to be ingested. Mechanisms regulating immune recognition and response to filamentous fungal pathogens are not well understood, although known risk factors for developing life-threatening infections are neutropenia or defects in the NADPH oxidase system. In this study, researchers showed human PMNs generate a respiratory burst response to unopsonized hyphae. Ab specific for beta-glucan, a major component of yeast cell walls, blocks this response, establishing beta-glucan as a key molecular pattern recognized by PMNs in response to C albicans. This study also elucidates recognition and signaling mechanisms used by PMNs in response to beta-glucan under conditions where phagocytosis cannot occur. Human PMNs adhered to immobilized beta-glucan and released an efficient plasma membrane respiratory burst. Ab blockade of the integrin complement receptor 3 (CD11b/CD18) significantly inhibited both of these functions. Furthermore, p38 MAPK and actin, but not protein kinase C. zeta, hasve a role in generating the respiratory burst to beta-glucan. Taken together, results show that beta-glucan in C albicans hyphae is accessible to PMNs and sufficient to support an innate immune response.
Potential applications: Biothera's yeast-derived, patented beta-glucan product, Wellmune, enhances immunity. It is marketed to manufacturers of dietary supplements, functional foods, cosmetics and animal nutrition.

Vital Stats: GTC's Nutraflora SCFOS
Study claims: Fructo-oligosaccharides (scFOS) are prebiotic ingredients that improve protection against pathogens, probably through promoting the growth of gastrointestinal bacterialike bifidobacteria and lactobacilli, which may lead to a better immune response.
Published: Karakan T, et al. Comparison of early enteral nutrition in severe acute pancreatitis with prebiotic fibre supplementation versus standard enteral solution: a prospective randomized double-blind study. World J Gastroenterol 2007 May 21;13(19):2733-7.
Abstract: Researchers compared the beneficial effects of early enteral nutrition (EN) with prebiotic fibre supplementation in 30 patients with severe acute pancreatitis (AP), who required stoppage of oral feeding for 48 hours, and who were randomly assigned to nasojejunal EN with or without prebiotics. The median duration of hospital stay was shorter in the study group [10 days (8-14 range) vs 15 days (7-26)]. Deaths occurred in six patients (20 per cent), two in the study group and four in the control group. Researchers concluded nasojejunal EN with prebiotic fibre supplementation in severe AP improves hospital stay, duration nutrition therapy, acute phase response and overall complications compared to standard EN therapy.
Published: Adogony V, et al. Effects of dietary scFOS on immunoglobulins in colostrums and milk of bitches. J Anim Physiol Anim Nutr (Berl) 2007 Jun;91(5-6):169-74.
Abstract: Researchers investigated whether supplementation with scFOS could enhance the mucosal immunoglobulin level in mammary secretions. Results showed that bitches supplemented with scFOS exhibited higher colostrum and milk IgM content without concomitant effect on IgG1, IgG2 and IgA.
Potential applications: NutraFlora scFOS prebiotic fibre supports immune health and digestive health and also improves calcium absorption.
